    Like the original series, "SPEC" will revolve around a pair of police detectives as part of a special division solving difficult cases (Mishou). Her partner Sebumi Takeru used to be the star performer of the Metropolitan Police. After some incident however, he was transferred to Mishou. Those two track down offenders with special abilities(SPEC) who managed to get away with their crimes.

    “IS” stands for “intersexual,” a term referring to people who cannot clearly be classified as male or female, and who may biologically possess characteristics of both sexes. Rokuhana's manga primarily deals with the troubles faced by the young Hoshino Haru (played by Fukuda), an intersexual who was registered as a girl at birth but has been raised as a boy.

    Goriki plays Aihara Miwako, a mysterious classmate who tries to get close to Haru. Inoue Masahiro has been cast as Ibuki Kenji, a member of the soccer club whom Haru begins having feelings for. Nishida Naomi and Minami Kaho play Miwako's and Haru's mothers, respectively.

Shun, Takeru and Kasumi (Watanabe Natsuna) with an equally troubled past and background, become collaborators after being hired by Kuchinawa (Kamikawa Tamaya) in police undercover operations in the city of 'Midori', a neighbourhood for illegal residents. They infiltrate a criminal gang to investigate, and find illicit human drug experiments and slave treatment staring them in the face. They grow to trust one another and mature through this experience.
