Brother's Keeper is a story about two brothers of the same mother but different fathers, "Sam" Kiu Tin-seng (Ruco Chan) and Lo Wai-son (Edwin Siu). Sam was born in Hong Kong, the son of Chow Yuk-mui (Louise Lee) and Kiu Sum (Lee Kwok Lun). Due to Kiu Sum refusing to marry Chow Yuk-mui, Yuk-mui decided to take Sam to Foshan, China to start anew. In Foshan, Yuk-mui got married with Lo Fu-shing (Lau Kong) and gave birth to Son a few years afterwards. Not long after, the family of four decided to cross the border to start a new life in Hong Kong. The family of four eventually got separated with Sam ending up in Hong Kong, while Yuk-mui, Fu-shing, and Son ending up in Macau. The two brothers lived very different lives in the course of the next 20 years. The very hardworking and ambitious Sam managed to graduate from college and became a police officer like his biological father, Kiu Sum. He has a girlfriend named Rachel Cheuk (Linda Chung) whom he is willing to do anything to fulfill her dreams of opening a fashion store. In order to achieve his goal, he seduces Fabio (Ankie Beilke) and uses her in the process. In addition, Sam also has an ambiguous relationship with a triad member named Keung Yung (Louis Cheung) who later caused Sam to end up in jail. Son, who is very simple-minded, only wants to live a peaceful and comfortable life in Macau. He is good friends with Yiu Man-ying (Krystal Tin) and Lung Fei (Louis Yuen). In order to avoid his abusive father (Fu-shing), Son works with his mother (Yuk-mui) at a restaurant. The lives of the two brothers, Sam and Son began to change after they meet up again. The two brothers in the series will go from 20 to 50 years old. This also shows the phrase "rags to riches" as Son becomes a rich businessowner from a poor young boy.

Sam writes books about crime-solving and murder, which Mon enjoys very much. What Sam does not know is that Mon has strong feelings for him. Mon is jealous of Ding Ding because Sam is dating her. But one particular day, the police force is called to a very strange case. The murder victims have been killed in the same way the victims were in Sam's books! When the force begins to suspect Mon - they find her dead. Who is this strange serial killer and why are they killing people following Sam's books?

After recovering from a long term illness, semi-retired popular singer Wah Fong Ying realizes that she has lived an unfulfilled life. With two failed marriages and estranged relationships with all of her children she lives a lonely life. Wanting to mend her relationship with her oldest daughter who resides in Taiwan, she travels there and meets Hong Kong photographer Ben Law, who is a fan of her. Fong Ying proceeds on working on her concert and hires assistant Jinny Szeto and creative director Sean Song to help her produce the concert. Just as light seems to shine on Fong Ying again, darkness comes back into her life when Jinny finds out that Fong Ying was involve in an affair with her uncle in the past and Ben starts to suffer from hallucinations due to a brain tumor.

She realizes that she is an incapable of taking care of Calvin, he needs Sunny more. Calvin takes her to meet his mom, but she has made her decision and breaks up with him. Despite this, Calvin is still in love with her, but they resume a professional attitude. The two of them eventually meet the new chairwoman of L'Ete Holdings, Catherine, who falls in love with Calvin. She knows that he is still in love with Sylvia but, more important, she knows how business-oriented the two were. Knowing that they wanted her because of her chairwoman position, she tells Sylvia to find a boyfriend and get married, only then will Catherine use her business to help Calvin.
